Research Says Reduce-Not Increase Antioxidants to Kill Pancreatic Cancer Cells.

Conventional wisdom in popular culture is to raise antioxidants in the body to facilitate recovery from cancer researchers have found that in pancreatic cancer the last thing one wants to do is raise antioxidants levels. Instead lowering antioxidants in pancreatic cancer cells can kill them them according to the researchers of the study published in  the journal Cell.  Pancreatic cancer is a lethal disease in which 5 per cent of the cases survive 5 years. /

In normal cells oxidizing and antioxidizing agents are created in every cell and kept in a precise balance. However, in proliferating cancer cells  more oxidizing cells are being made in malignant cells but more antioxidants are being made also that counter the impact of rising oxidation and without commensurately more antioxidants the  malignant cells will die due to excessive oxidation and that is what is desired. Researchers tested  whether by increasing the level of oxidation in cancer cells,the pre-cancerous cells and malignant cells would die. When cells detect excessive oxidation, they commit suicide, following a built in program called apoptosis and one way to increase oxidation in cancer cells is to decrease levels of antioxidants in those same cells.

Researchers were interested in how best to do this without harming normal cells. The focused on a protein called NRF2 which is considered a master regulator of redox homeostasis or by which they can turn the switch to tweak and disturb the exquisite balance between oxidation and reduction in cancer cells. When NRF2 is active, cells synthesize a chemical called glutathione that is an important antioxidant. Thus, by reducing NRF2 activity or knock it out of action would seem desirable but this is not possible for two reasons. First, it is a transcription factor—a protein that regulates the activities of other genes.  Transcription factors is famously difficult to target but researchers say that would not be desirable because in addition to promoting  production of glutathione, NFR2 has a role in regulating several hundred different gene that impact on many other processes.

After conducting experiments and testing pancreatic cells in animal models they arrived at a different strategy. They used a panel of pancreatic organoids,  spherical agglomerations of pancreatic cells, sampled from people with pancreatic cancer and from healthy pancreas and were able to observe what happens when NRF2 is completely eliminated. They ran the test on normal, premalignant and malignant pancreatic organoids. They saw that the premalignant organoids carried cellular mutations in the Kras gene that is aberrant in almost all human pancreatic cancers. The malignant organoidshad that mutation and also a mutation that inactivates the powerful tumor supressor gene P53 and these with Kras are seen in most human malignancies.

Researchers, thus, saw that when NRF2 is missing the mechanism in cells that translate messages from genes to proteins (protein synthesis) is very sensitive to fluctuations in the balance between oxidants and anti-oxidants. However, protein synthesis was not impacted in normal pancreatic cells. Researchers said “We were very excited when we saw this. This meant that if we could find a way of reducing antioxidants, protein synthesis would only be impacted in precancerous and malignant cells, a potentially powerful therapeutic strategy.”

Using a mouse model they found that using two drugs–one that inhibited the beginning of the translation process that leads to protein synthesis and the other inhibited the synthesis of glutathione which is an antioxidant- was more effective in killing cancer cells that either alone and normal pancreatic cells were not affected. Further research to look at combinations and to increase efficacy are planned.

Do Antioxidants Cause Malignant Melanoma to Metastasize

Logos 005A new study published in Science Translation Medicine concluded that antioxidants can double the rate of melanoma metastasis in mice and reinforces previous research that antioxidants hasten the progression of lung cancer.  Researchers further said that those with cancer or the high risk of cancer should avoid   nutritional supplements that contain antioxidants. Because it was known that free radicals can cause cancer, the research community assumed that antioxidants that destroy them, provided protection against cancer. As a result many nutritional supplements included antioxidants as a cancer prevention.

Now research shows this not to be true. The current study on melanoma, the worst type of skin cancer, found that antioxidants doubled the rate of metastasis in malignant melanoma and as opposed to the lung cancer study the primary melanoma tumor was not affected. Researchers said “We have demonstrated that antioxidants promote the progression of cancer in at least two different ways.” They protect healthy cells from free radicals that can turn into malignancies but may also protect a tumor once it has developed. Thus, taking nutritional supplements containing antioxidants may unintentionally hasten the progression of a small tumor or premalignant tumor, neither of which is possible to detect. Thus, they say those recently diagnosed with cancer should avoid antioxidant nutritional supplements.

The role of antioxidants is particularily relevant with patients who have melanoma because melanoma cancer cells are know to be sensitive to free radicals and alsoi because the cells can be exposed to antioxidants by non-dietary means . “Skin and suntan lotions sometimes contain beta carotine or vitamin E, both of which could potentially affect malignant melanoma cells in the same way as antioxidants in nutritional supplements.”  The role of lotions on cancer and the effect of antioxidants on cancer are ongoing in research by this orgainzation.

NB:It is interesting that research published April 9, 2014 on this site concluded that vitamin A (beta carotine) prevented normal cells from reverting to cancer cells.

Are Antioxidants useful for Improving the Immune System of the Elderly?

Antioxidants and Immunity: A new study published in the journal Cell Reports concluded aging cripples the production of new immune cells and decreasing the immune systems response to vaccines and putting the elderly at risk of infections. They also stated that antioxidants in the diet slow this damaging effect.

Researchers focused on the thymus that produces T lymphocytes that are critical immune cells that must  be continuously replenished in order for the body to respond to infections. Unfortunately this organ begins to atrophies rapidly in early adulthood  and looses its function. In order to understand the process of how antioxidants slows the process of thymus atrophy and T lymphocyte production they analyzed the activities of genes in two major thymus cell types–stomal cells and lymphoid cells—in mouse tissue and found that stomal cells were specifically deficient in an antioxidant enzyme called catalase, which results in elevated levels of the reactive oxygen by-product of metabolism and, subsequently accelerates metabolic damage.

To confirm  the central role of catalase, researchers increased the levels of this enzyme in genetically altered animal models, resulting in the preservation  of the size of the thymus for much longer. In addition, animals that were given two common dietary antioxidants including vitamin C were also protected from the effects of agin on the thymus. These results take together support the free radical theory of aging that proposes that reactive oxygen species such as hydrogen-peroxide, that is produced during normal metabolism, causes cellular damage that contributed to aging and age-related diseases.

Can Certain Flavonoids Kill Human Pancreatic Cancer Cells?

logo1267406_mdA new study published in Molecular Nutrition and Food Research concluded that celery, artichokes, and herbs, especially Mexican oregano, all contain apigenin and luteolin, flavonoids that kill human pancreatic cancer cells in the lab by inhibiting an important enzyme. They found that apigenin alone induced cell death in two aggressive pancreatic cancer cell lines but the best results were received when the cancer cells were pretreated for 24 hours with apigenin and then applied chemotherapeutic drug gemcitabine for 36 hours.

Thus, using the flavonoids as a pre-treatment instead of applying it simultaneously with the chemotherapy drug worked best. The author said “Even though the topic is still controversial, our study indicates that taking antioxidant supplements on the same day as chemotherapeutic drugs may negate the effect of those drugs.” “That happens because flavonoids can act as antioxidants. One of the ways that chemotherapeutic drugs kill cells is based upon their pro-oxidant activity, meaning that flavonoids and chemotherapeutic drugs may compete with each other when they are introduced at the same time.”

Researchers found that apigenin inhibits an enzyme called glycogen synthase kinase-3β (GSK-3β) that led to a decrease in the production of anti-apoptotic genes (self-destructing) in the pancreatic cancer cells. The percentage of cells undergoing apoptosis went from 8.4 percent in cells that had not been treated with the flavonoids to 43,8 percent in cells that had been treated with 50-micromolar dose. In this case, no chemotherapy drug had been added.  Treatment with the flavonoids also modified gene expression especially those associated with pro-inflammatory cytokines.

He then went to Brookhaven National Laboratory for Post-doctoral training. Later, he was appointed as the Director for the Center for Vitamins and Cancer Research. He has written several book chapters and abstracts as well as authored or edited 18 books on radiobiology, radiation protection and nutrition and cancer. He is a member of several professional organizations, and serves as an ad-hoc member of various Study Sections of the National Institute of Health (NIH). He is a frequently invited speaker at National and International meetings on nutrition and cancer. In 1982, he was invited by the Nobel Prize Committee to nominate a candidate for the Nobel Prize in Medicine. He was selected to deliver the 1999 Harold Harper Lecture at the meeting of the American College of Advancement of Medicine. He is a former President of the International Society for Nutrition and Cancer.  Dr. Prasad has consistently obtained NIH grants for his research. His current interests are in the area of radiation protection, nutrition and cancer and nutrition and neurological diseases, particularly Alzheimer’s disease and Parkinson’s disease.
